A trait reroll swaps a unit's current trait for a new random one. You only need one reroll per use no matter how rare the unit is, and using another reroll overwrites whatever trait the unit already had.

What a trait actually changes
Traits are bonuses attached to a unit that can boost its stats or abilities, sometimes by a huge margin. The better the bonus, the lower its drop rate, with some of the strongest traits landing as low as a 0.1 percent chance per reroll. A unit can only hold one trait at a time.
How to reroll a trait
Find the Mandra NPC in the lobby's Upgrades area. Interact with Mandra, pick the unit you want to reroll, usually your rarer units first since a good trait matters more on them, and press the reroll button. You can also get lucky and pull a unit that already has a strong trait straight from a summon.
Farming rerolls for free
You do not need to spend Robux to build up a stock of rerolls. They come from completing achievements and challenges, hitting level milestones, working through the Battle Pass, and from codes like MiniUpd2 and Retribution. Stack up a handful before you start rerolling so a string of bad pulls does not leave you stuck.
